Nitrofurantoinの神経毒性に関する実験的研究

Neurotoxicity with nitrofurantoin. An experimental study

Ⅰ．緒言

　キノホルム以来，薬剤によるpolyneuropathyが注目されている。尿路感染症に対して用いられるNitrofurantoin（Furadantin^®，以下NFと略す）によると思われるpolyneuropathyは1956年以来，報告があり^12），1973年までの報告例は欧米で137例にのぼるといわれ^18），わが国でも5例の報告がある^{2,3,6,10,11）}。一方，動物実験ではラットで神経症状がみられたにすぎない^1,7）。

　われわれは，最近NF-plolyneuropathyの臨床例を経験したのを機会に，イヌに対して本剤を投与しneuromyelopathyの所見を得たので報告する。

　In an experiment to disclose neurotoxicity of nitrofurantoin and to compare it with those of other drugs which induce neuropathies, the drug was administered orally to five dogs. Three dogs which were given relatively large doses of 67~100 mg/kg/day died on the 8 th, 15 th and 17 th days. Other two dogs were administered smaller doses of 20~50 mg/kg/day for 107 and 153 days, respectively.

　The dog which was given the substance for 107 days showed muscle weakness in legs and ataxia such as side-swaying of hips and wide-opening of hindlegs.
