　Charcot-Marie-Tooth病（CMT）23例を対象にPMP−22遺伝子の重複を，3つ方法：1）PMP—22cDNAを用いたサザン法；2）重複部位を認識するpVAW409R3を用いたサザン法；3）重複部位を認識するマーカー6G1部位のPCR法，により検索するとともに，重複の検出におけるこれらの方法の有用性を対比した。pVAW409R3により検出される高分子DNAのMspI切断断片，2.8kbと2.7 kbのバンドの比は重複例では1.75〜2.13，非重複例および健常例では1.01〜1.15に分布した。また，BamHI切断後のPMP—22cDNAとSF85（第21染色体上の内部コントロール）による検討では，その比は健常例を1とする非重複例では0.96〜1.04，重複例では1.15〜1.33であった。一方，6G1とD1S80 locus（第1染色体上の内部コントロール〉との定量的PCR法ではその比は健常例と非重複例では1.29〜1.74，重複例では1.67〜2.23に分布し一部両者にoverlapがみられた。PMP−22遺伝子の重複の検出にはpVAW409R3によるサザン法が比較的容易であるが，PMP−22 cDNAによる方法が必須と思われた。また，CMT type 1の大部分（80％）にはPMP—22遺伝子の重複が存在し，従来の欧米からの報告を確認した。

We investigated duplication of the PMP-22 gene in 23 Japanese patients with Charcot-Marie-Tooth disease (CMT) by Southern blot and PCR analysis. To detect duplication of the PMP-22 gene region, PMP - 22 cDNA and a polymorphic marker, VAW409R3, located in the region flanking the PMP-22 gene, were used as probes for Southern-blot analysis. A marker, 6G1, also located in the PMP-22 flanking region, was amplified using the quantitative PCR method.
