近年,miRNAをはじめとするnon-coding RNAや選択的スプライシングの制御が,生物の発生や病態へ関与するとともに,臨床診断における分子マーカーとしても注目されつつある.そのため,より正確な蛋白質―RNA相互作用の検出技術,およびそのゲノムワイドで包括的なRNA制御機構の理解が必須となってくる.そこで今回,HITS-CLIP法(high through put sequencing UV cross linked immunoprecipitation)による蛋白質―RNA相互作用検出方法を紹介し,今後の臨床診断への応用についても議論してみたい.
An increasing number of studies have demonstrated that the complexity of mammalian transcriptome accomplished by RNA processing and miRNA regulation are involved in developmental mechanisms and pathophysiologies of diseases. Especially, one set of miRNAs is thought to be a new class of biomarkers in cancer. Therefore, bona-fide mapping of protein-RNA interactions in the genome would be necessary for a comprehensive understanding of RNA regulation in normal physiology and pathology. Here I introduce an unbiased HITS-CLIP technology to detect in vivo protein-RNA interactions and discuss the possibility of application in clinical diagnosis.
