Circulating mRNA and microRNA as biomarker

末梢血中を循環する核酸は，新たなバイオマーカーとしてこれまで注目されてきた．しかしその存在様式などは長らく不明であった．最近mRNAや小分子非翻訳RNAであるマイクロRNA(microRNA；miRNA)が，小胞顆粒であるエクソソーム中に存在することが明らかとなり，血中に存在するmRNAやmiRNAを用いたバイオマーカーの探索が再注目されている．これらの核酸はエクソソームを始めとする顆粒や蛋白質に結合しているため体液中で安定であるが，その解析技術に関してはまだ不十分である．しかしPCRやマイクロアレイ，次世代シークエンサーなどの新規の技術発展により今後の臨床応用が期待できる．

Serum and other body fluids are rich resources for the identification of novel biomarkers.　Recently, it was discovered that extracellular microRNA (miRNAs) and mRNA circulate in the blood of both healthy and diseased patients.　Most of the circulating miRNAs and mRNA are included in protein, lipid or lipoprotein complexes, such as RNA-binding proteins, apoptotic bodies, microvesicles, or exosomes, and are, therefore, highly stable.　The existence of circulating miRNAs and mRNA in the blood of cancer patients has raised the possibility that circulating RNAs may serve as a novel diagnostic marker.
