Immune mediated myelin destruction and thedemyelination secondary to viral infection of the oligodendroglia are well accepted mechanisms of virus induced demyelination. Demyelination in the CNS may occur as a consequence of systemic viral infections and vaccinations where the host immune mechanism plays a decisive role and the evidence of direct virus invasion into the CNS is often lacking. Progressive multifocal leukoencephalopathy is the only human demyelinating disease in which demyelination is directly related to the viral infection of myelin supporting oligodendroglia. In addition, causative role of viral infections has been postulated in a variety of human demyelinating diseases, including multiple sclerosis, but no convincing evidence has been provided.
