輓近に於ける2, 4-Dinitrophenolに関する研究の動向

　2,4-Dinitrophenol（DNP）並びにその近縁物質が甚だ特異的な薬理学的並びに生化学的能動性を有することは，1918年以降約20年の間にMagne^1），Tainter ＆ Cutting^2）〜5），Heymans^6），Krahl ＆ Clowes^7）〜10），Ronzoni ＆ Ehrenfest^11）等により，その概要が明らかにされた。ところが1948年Loomis ＆ Lipmann^12）によりDNPが酸化的燐酸代謝を特異的に分離阻害することが立証され，上記の諸氏により記載された本剤の作用は全てこのuncoupling Propertyを根底として惹起されるものと推量されるに至つた。更に近時急速に発展した酵素学の進歩と相俟つて，生化学的諸反応或いは生体諸機能の発現乃至維持が如何程力源授受に依存するかを闡明する好個の解析物質として本剤が盛んに応用されるに至つた。而して過去に得られたDNPの基本的性格に関しては，既にEdsall^13），Simon^14）及びBrody^15）の綜説に記載されているので，私はこれらの綜説事項と重複する点はその概要を述べるにとどめ，むしろ過去に於て比較的等閑視された本剤の代謝状況や最近得られた生体内諸代謝への影響に関する知見等を中心として論述する。

　DNP produces the marked carbohydrate consumption associated with the stimulation of respiration in aerobic system in vitro, and these phenomena have been explained by the specific effect of the drug which interrupts the normal linkage of oxidation and phosphorylation.

　Although the responses to DNP in vivo are similar outwardly to those in vitro, we should rather consider the participation of some endocrine glands, especially adrenals.

　On the other hand, the fact that DNP is readily inactivated (reduction or conjugation) by living cells should be noted to discuss the mechanism of actions of DNP.
