Dyskinesia produced by l-dopa in monkeys with resting tremor and tegmental lesions

I．はじめに

　パーキンソニズムに対するL-Dopaの効果が確立されてより，本症候群は神経化学の面よりstriatal dopamine deficiency syndrome（Hornykiewicz，1973）と呼ばれるに至っている。しかし，L-Dopa療法は大量投与の必要があるため，副作用が多く，L-Dopaの効果を高め副作用を少なくする目的で，monoamine xidase抑制剤（Barbeau，1969），catechol-O-methyl transferase抑制剤（Ericsson，1971，Kremznerら，1973），dopamine-β-hydroxylase抑制剤（Menaら，1971）などの酵素抑制剤の併用のほか3-O-methyl dopa（Bartholiniら，1971）やドーパミン受容体の刺激剤（Corrodiら，1972）とされているPiribedil（ET 495）の効果も検討されたが（Chaseら，1974），いずれも満足すべき効果をうるに至っていない。脳内ドーパミン（DA）を高めるためには，L-Dopaと末梢性脱炭酸酵素抑制剤の併用が最も効果的であることがわかり（Porterら，1962），臨床応用も試みられている（Mardsenら，1973）。

Resting tremor of the unilateral extremities was produced in African green monkeys (Cercopithecus subaeus) by a lesion in the contralateral ventromedial tegmental area of the brain. The coordinates of the ventromedial tegmental area were 1.5~2mm from the sagittal plane, 8~10 mm above the interaural line, and 7~9mm anterior to the interaural line. Hypokinesia of the contralateral extremities appeared immediately after placement of a radio-frequency lesion in the ventromedial tegmental area.
