Methylnitrosourea慢性経静脈投与によるラット神経腫瘍，脳・脊髄腫瘍の実験病理学的研究

Morphological studies on the methylnitrosourea-induced tumors of the nervous system, with special reference to nerve tumors

I．はじめに

　脳脊髄神経に発生する神経腫瘍の組織由来については現在なお定説がなく，さまざまな名称で呼ばれてきた。Masson^20）（1932），Stout^27）（1935）らはこの神経腫瘍の組織由来をSchwann細胞と考え"Schwannoma"，"Neurilemoma"とし，近年Russell ＆ Rubinstein^25）（1971）も同様の見解を示している。一方perineurial connective tissue由来と考えるMallory^21）（1920），Penfield^22）（1927），Tarlov^29）（1940），らは"Perineurial fibroblastoma"と呼び，Zimmerman et al．^32）（1956）は結合織染色の反応を示すこの腫瘍を便宜的にperineurial fibroblastomaと呼んでいる。最近，この腫瘍の電顕的観察が行なわれるようになったが，腫瘍細胞の起源についてはなお意見が分かれている。

　化学発癌物質による脳腫瘍の実験的研究は主として成熟動物の脳内にpellet状の発癌物質を挿入する方法で行なわれてきた。この方法によって形成された実験的膠腫は発癌物質を中心に形成されており，組織学的に悪性の成熟型膠腫であることが確かめられてきている^13）。

A total of 103 Donryu rats at the age of 3 or 4 weeks, weighing 30 to 40 g, were used and divided into 3 groups. The animals in Group A were weekly given intravenously 10 mg/kg of methylnitrosourea (MNU) dissolved in saline, and animals in Group B 5 mg/kg of MNU. In Group C, control rats were weekly treated intravenously with saline.

Fifty-four rats out of 79 given MNU, with duration of survival ranging from 137 to 365 days, were found with single or multiple tumor development in the nervous systems. The number of the tumors induced amounted to 73.
