Abstract 文献概要

　補体抑制薬C1 esterase inhibitor（C1 INH）を用いて再灌流傷害抑制効果を検討した。

　〔方法〕29匹のWistar ratを用いた。栓子法で60分間の中大脳動脈閉塞を行い，再灌流直前にC1 INH 50IU／kg（C1 INH群：n＝15）または生理食塩水（対照群：n＝14）を静脈内投与した。実験中は血圧，心拍数，体温，局所脳血流，および動脈血液ガスデータをモニターした。48時間の再灌流後に梗塞体積およびmyeloperoxidase（MPO）活性を測定した。

　〔結果〕血圧，心拍数，局所脳血流は両群で有意差は認められなかった。梗塞体積およびMPO活性は，対照群（179.5±92.8mm³，1.27±0.46U／g）と比較しC1 INH群（86.5±76.8mm³，0.38±0.30U／g）で有意に減少した（p＜0.01）。

　〔結論〕C1 INHを用いた補体活性抑制は，虚血巣内への白血球浸潤を減少させ再灌流傷害を抑制することが証明された。

〔Background〕Despite the current interest in thrombolytic therapy for acute stroke, ischemia-reper-fusion injury remains a potentially hazardous compli-cation. The complement system is thought to play a major role in initiating some of the inflammatory events occurring in the reperfusion injury. This study was conducted to explore the effect of C1 esterase in-hibitor (C1 INH) on the reperfusion injury in rat mid-dle cerebral artery (MCA) occlusion-reperfusion model. (Methods) Twenty-nine male Wistar rats were used. Intraluminal MCA occlusion was performed for 60 minutes.
