　今回我々は，グリオーマ細胞からのG-CSFの分泌の有無ならびにG-CSFのグリオーマ細胞の増殖能に対する影響を調べた。培養細胞上清をELISAにて調べたところ，8種のグリオーマ細胞株中3種の細胞株でG-CSFが検出でき，3種においてはTNF—α刺激でG-CSFの発現が認められた。これをmRNAレベルで確認するためにRT-PCRを行ったが，ELISAにてG-CSFが検出できなかった細胞株においてもG—CSF mRNAが検出し得た。G-CSFの脳腫瘍細胞の増殖能に対する影響をみるためにproliferation assayを行ったが，G-CSFはグリオーマ細胞の増殖能に影響を与えず，RT-PCRにてグリオーマ細胞にG-CSF受容体は検出し得なかった。以上より，多くのグリオーマ細胞はG-CSFを分泌するが，その受容体を欠くために，autocrineとしての作用はないものと思われた。

In this study, we investigated the expression of granulocyte colony-stimulating factor (G-CSF), G-CSF mRNA, G-CSF receptor mRNA in glioma cell lines, G-CSF in glioma cyst fluids, and the effect of recombinant G-CSF on the proliferation of glioma cells. First, to determine whether G-CSF is pro-duced by glioma cells, we analyzed for the presence of G-CSF by ELISA in supernatants from glioma cell lines. G-CSF was detected in six of fourteen glioma cell lines constitutively, and, after stimula-tion with tumor necrosis factor-α (TNF-α), G-CSF was detected in four of eight cell lines which did not produce G-CSF constitutively.
