An accurate detection of the cerebral amyloid deposition is important in the clinical drug trial focused on the early stage of Alzheimer’s disease. A less invasive and cost-effective blood-based biomarker is desired for large-scale screening. We developed a high-sensitive assay for plasma Aβs using a combination of immunoprecipitation and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. In a collaborative research with National Center for Geriatrics and Gerontology, we discovered an effective blood-based biomarker(APP669-711/Aß1-42)for cerebral amyloid deposition. Subsequently, a pilot study led to the development of a “composite biomarker,” which combined APP669-711/Aß1-42 with Aß1-40/Aß1-42. An analysis of plasma samples from 373 subjects in Japan and Australia showed that our biomarker could predict the presence of abnormal Aβ deposition with high accuracy. Our assay is useful for early screening of amyloid-positive subjects and could be expected to contribute to the advancement of drug discovery.
