Vitamin D3 is metabolized to 25(OH)D3 in the liver by CYP2R1 and subsequently metabolized to 1α, 25(OH)2D3 in the kidneys by CYP27B1 before exerting its biological action. In target cells, CYP24A1 is highly expressed for inactivating 1α, 25(OH)2D3 soon after expressing biological activity. The activating mechanism and inactivating mechanism of vitamin D3 by the enzymes are regulated by many factors involved in keeping bone mineral homeostasis. Elucidation of overall vitamin D3 metabolism is required to clarify the mechanism of the action of vitamin D3.
