The major circulating form of vitamin D metabolites in serum is 1α,25-dihydroxyvitamin D3 and 25-hydroxyvitamin D3. 1α,25-Dihydroxyvitamin D3 is measured for the evaluation of calcium metabolism, and 25-hydroxyvitamin D3 is measured for the evaluation of vitamin D nutritional status. The assay variation present in measurements of 25-hydroxyvitamin D3 confounds the diagnosis of hypovitaminosis D, and requires standardization from the clinical and nutritional standpoints. Because the National Institute of Standards and Technology (NIST) developed a Standard Reference Material 972 (SRM972), we introduce in this review a standardized procedure for calibration of serum 25-hydroxyvitamin D3 measurements using SRM972.
