PC12細胞由来新規神経栄養因子

Neurotrophic factors derived from PC12 cells.

ラット褐色細胞由来のPC12細胞は，神経分化ならびに細胞死のモデルとして様々な研究に用いられてきた。私達は同細胞にヒトbcl-2遺伝子を導入したPC12細胞（bcl-2／PC12細胞）を樹立し，同細胞が無血清培養下で生存するだけではなく，突起を伸展して神経細胞様に分化することを示した。さらにこの細胞の培養上清は野生型PC12細胞のアポトーシスを回避させ，突起伸展活性を示した。この生存活性を指標として，因子の同定を試みたところ35kDの分泌性の糖蛋白を見出した。本蛋白の解析から，胎生期の神経上皮細胞に発現すること，神経細胞の分化・生存活性を有すること，lesion factorとして作用する可能性があること，などが明らかとなってきた。

PC12 cells, a rat adrenal pheochromocytoma cell line, are known to grow when cultured in the presence of serum, while they extend their processes and differentiate into neuron-like cells when cultured in the absence of serum but in the presence of nerve growth factor (NGF) . The cells have also been shown to undergo apoptosis following serum/trophic factor deprivation. Therefore, the cells have been used as a model of neuronal differentiation and apoptosis in vitro. There are many subpopulations in PC12 cell lines which show different natures.
