A pivotal role of neurotrophic factors and their receptors in neural cell survival and differentiation has been well established especially from the studies of knockout mice of these genes. Among PNC, severe neuron loss was observed from superior cervical ganglion and DRG in NGF/trkA knockout mice and from vestibular in BDNF/trkB knockout. Neurotrophic factors activate Trk receptors tyrosine kinases, thus trigger ras/raf-1/MAPK cascade. This cascade induces accumulation of bcl-2 protein, which is a negative regulator of apoptotic cell death.
