はじめに

　1959年にCarlsson^1）が黒質・線条体路がドーパミンであることを見出し，レセルピン投与によりパーキンソン病様の運動減少を引き起こすことを見出して，最初のパーキンソン病モデル動物を作成した。このことは，それ以後に展開されたL-DOPA療法のはなばなしい歴史をつくり上げるきっかけをつくった。

　6-ハイドロキシドーパミン（6-OHDA；6-OH-dopamine）がUngerstedt^25）により1971年に導入され，主として一側のドーパミンニューロンを破壊するのに成功し，その後長い間，パーキンソン病のモデルとして使用され，さまざまな薬理学的研究に使われ，新薬開発など大きな貢献をしてきた。しかし，あえて6-OHDAのモデルとしての欠点をあげると，主として6-OHDAが使われたのはラットであり，かつまた一側性の障害モデルでしかなかった。つまり両側の黒質ドーパミンニューロンを破壊するとラットは死亡する。また6-OHDAは血液・脳関門を通らないため，腹腔，静注，皮下注などの全身投与では脳内に到達しない。そのため，黒質・線条体路などに直接注入するか，脳室内投与などの方法がとられてきた。またサルでの6-OHDAモデルはあまりつくられていない。

　The fact that the use of MPTP produced symptoms similar to Parkinson's disease led to the hypothesis that the substances which had similar chemical structures to MPTP and neurotoxicity to the nigro-striatal dopamine neuron, and existed within the brain or in nature might cause the disease.

　The mechanism of cell death of the dopamine neuron by MPTP may possibly be due to energy crisis by the inhibiting effect of mitochondrial complex I activity by MPP⁺.
