ミトコンドリア病による知的発達障害の分子遺伝学

Molecular genetics of mental developmental disorders caused by mitochondrial disease

　ミトコンドリアの機能障害によるミトコンドリア病において，知的発達障害をきたす臨床病型が知られている。その代表的な臨床病型であるLeigh脳症とMELASを取り上げ，その分子遺伝学的な知見を紹介する。ミトコンドリア病の病因は，核DNA上の遺伝子変異とともに，ミトコンドリア内に存在するミトコンドリアDNAの異常に起因する場合がある。ミトコンドリア機能異常に起因する知的発達障害の発生機序の解明は，その大部分は今後の研究に依存している。

　It is well known that there are clinical phenotypes with mental developmental disorders in mitochondrial disease. Molecular genetics of two representative entities of Leigh encephalopathy and MELAS（mitochondrial myopathy, encephalopathy, lactic acidosis and stroke-like episodes）is described. Both nuclear gene defects and mitochondrial DNA abnormalities are the causes of mitochondrial disease. Pathogenesis of mental developmental disorder due to mitochondrial dysfunction depends heavily on the future study.
