 In April 2000, we started clinical trial of interferon-β(IFN-β)gene therapy for the patients with malignant glioma. As a vector we used our original cationic liposomes, which are multilamellar typed liposomes. Treatment consists of reoperation, injection of liposomes containing human IFN-βgene on day 0, 14, 21, and 28, end of treatment evaluation on 3months later from the first day of liposome administration. In patients undergoing tumor resection, the surgical margin of the cavity was infiltrated with 1ml of liposomes containing human IFN-βgene at a concentration of 30μg DNA/ml, evenly distributed at multiple sites. From 2 to 4 injections, repeated procedure was stereotactically performed under local anesthesia. Safety and tolerability was assessed by physical examination, vital signs, laboratory evaluations, adverse events and concomitant medication usage. Transgene expression and antitumor activity were detected. Two patients showed a partial response(>50%tumor reduction)and others had stable disease at 3months after the first injection. The study suggests feasibility and safety of IFN-βgene therapy, which may become an important treatment option for patients with malignant glioma.
