Polymerase Chain Reaction（PCR）法によるLeber病の早期診断の試み

Early diagnosis of Leber's optic neuropathy by polymerase chain reaction

　臨床的にLeber病が強く疑われた34歳と32歳の兄弟と眼科的に正常の妹の末梢血の白血球からミトコンドリアDNAを抽出して11778番塩基対の検討を　PCR （Polymerase Chain　Reaction）法を使って行った。この三人には正常者にはみられない11778番塩基対の点突然変異を認めLeber病の家系と考えられた。この方法はLeber病の診断に非常に有用と考えられた。

We diagnosed a pair of brothers, aged 32 and 34years, as presumed Leber's optic neuropathy We analyzed the mitochondrial DNA from the leuco-cytes as to the nucleotide position nt 11778 using polymerase chain reaction. Both cases and their apparently healthy sister showed a point mutation in nt 11778. This method was thus useful in establi-shing the diagnosis of the disease.
