Abstract 文献概要

　一医療施設で治療中の結膜炎患者の瞼結膜から，経日的に採取された7例の患者を含む結膜材料22検体，およびトラコーマ群の28例の眼結膜擦過材料28検体についてPCR法増幅を試みた。回収液にSDS-ETを使用した結膜炎患者の擦過材料では，螢光抗体法を応用した市販のMicro Trak®で抗原陽性と判定された4例の患者の18検体のうち11検体からクラミジア遺伝子が検出された。また治療中，抗原陰性と判定された日に採取された9検体のうち，2検体がクラミジア遺伝子陽性であった。30サイクルと40サイクルの増幅成績の比較では，治療中の同一患者の検体においてDNA増幅量の経日的な減少が示され，治療との対応が認められた。また，同時に供試されたクラミジア抗原陰性患者の瞼結膜擦過材料のDNAと鋳型DNAを含まない反応液のみの対照群において増幅はみられなかった。クラミジア抗原陰性で血中抗体価の高かったトラコーマ患者でのPCR増幅はすべて陰性であった。

Polymerase Chain Reaction (PCR) technique was applied for the diagnosis and epidemiological study of Chlamydiosis. The aim of this study was to establish the basic technique of PCR for the sensi-tive and specific detection of Chlamydia and to apply PCR for epidemiological survey and diagno-sis of human chlamydiosis.

PCR was also performed on human clinical speci-mens including 4 cases of conjunctivitis (18 swabs from conjunctivas), 28 cases of trachoma (28 swabs from conjunctivas), and 3 cases of normal controls (3 swabs from conjunctivas).

Out of 18 samples from conjunctivitis patients positive for antigen with immunofluorescence assay (IFA), 9 were positive in PCR. Out of 9 conjunctiva samples negative for antigen, 2 was positive in PCR. The samples of trachoma were all negative in both antigen and PCR.
