We used the technique of DNA amplification with the polymerase chain reaction to detect the virus genome in ocular samples from a patient with clinically diagnosed acute retinal necrosis (ARN) . Using VZV primers, we detected the VZV genome in the aqueous humor, but not in the vitreous, from the patient, amplifying a DNA fragment 642 base pairs in length. After detection of the VZV genome, we used the Pst I restriction endonuclease for VZV strain differentiation because an epidemiological study found the Pst I -site-less mutation in 25% of the VZV strains seen in Japan. The VZV genome from the patient had a Pst I cleavage pattern, but positive control represented a VZV carried a Pst I -site-less mutation. From the present study the patient was thought to be caused by infection of VZV having a Pst I cleavage pattern.
