  • Look Inside

We studied the binding of seven different kinds of lectins to the retinoblastoma of 4 eyes and com-pared with that to the normal human and rabbit retinas. Peroxidase labelling method was used forlight and electron microscopic observation of lectin binding pattern.

In the normal rabbit retina, wheat germ agglu-tinin (WGA) stained the rod and cone layer and the outer nuclear layer. Peanut agglutinin (PNA) stained the rod and cone layer and the outer nuclear layer. Ricinus communis agglutinin (RCA) stained the outer nuclear layer, the inner limiting layer, and the nerve fiber layer. Concanavalin A (ConA) stained all the layers of the retina.

In the human retina, WGA stained the rod and cone layer, whereas ConA stained all of the layers.

In the human retinoblastoma, ConA stained the cytoplasm and the cell membrance of both the differentiated and the undifferentiated types. By electron microscopy, the cell membrane, the nuclear envelope, and the endoplasmic reticulum were stained. WGA stained the cell membrane of the cells of both types. A particularly strong staining was observed in the central part of the Flexner-Wintersteiner rosette formation. These results sug-gest that the retinoblastoma is similar in structure to the photoreceptor cells in terms of the saccharide components.
