網膜静脈血栓症に対する合成抗血液凝固剤Dextran Sulphateの効果について

Anticoagulant treatment of retinal vein thrombosis with a new synthetic anticoagulant Dextran Sulphate

　動物組織（主に肺・肝）より抽出精製されるHeparinは優れた抗血液凝固剤として臨床的に広く用いられているが，高価の点が使用上の隘路である。

　Jorpes （1935）によりHeparinはMucoitinpolysulphuric acidの一種でGlucuronic acidとAcetylglucosamineからなる高分子硫酸エステルであることが明らかにされて以来，Heparinと同様の作用を有つ硫酸エステルを合成によつて得ようとする試みが数多くなされて来た。その結果澱粉，グリコーゲン，キチン，セルローズ，イヌリン及びその他種々の多糖類の硫酸エスエルが合成され，予期した抗凝血作用は得られたが，毒性が強くて臨床的に使用されるには至らなかつた。

Synthetic anticoagulant, Dextran Sulphate was administered for 7 to 10 days ; on the first day by four times intravenous injections, on the second day thrice, and thereafter once or twice daily intermittently. The dose of injection was kept each time at 300mg. The frequency and interval of injections were controlled by determinaing clotting time as an indicator.

A cumulative effect in clotting time was observed after the repeated injections. There was a marked fall in serum total cholesterol and platelet count during the treatment.
