I．はじめに

　オピオイドベプチドはモルヒネ・レセプターに結合し，モルヒネと同様の鎮痛効果を現わすペプチドの総称である。1975年，Hughesら^1）によって2個のpentapeptides，methionine⁵-enkephalin（Met-enkephalin）およびleucine⁵-enkephalin（Leu-enkephalin）が見出されて以来，現在までに多数のオピオイドペプチドが発見された。これらペプチドはその前駆体から，現在，3群に分けることができる（表1）。第1群は，ACTHと共通の前駆体，proACTH-β-LPH（proopiomelanocortin）より由来すると考えられるペプチドで，β-endorphinがその代表である。第2群は，proenkephalin Aに由来するペプチドで，Met-enkephalin，Leu-enkephalin，Met-enkephalin-Arg⁶-Phe⁷，Met-enkephalin-Arg⁶-Gly⁷-Leu⁸，およびこれらペプチドを含む大分子前駆体（peptide E，F，A，Bなど）がこれらに含まれる。第3群オピオイドペブチドは，proenkephalin B（prodynorphin）に由来するもので，N端にLeu-enkephalin構造を有しており，dynorphinやα-，β-neoendorphinがこれに含まれる。

　Opioid peptides in the body can be groupedinto 3 categories. The first group consists of peptides derived from proACTH/β-LPH (proopiomelanocortin) and β-endorphin represents this group. The second group contains peptides such as Met-enkephalin, Leu-enkephalin, Met-enkephalin-Arg⁶-Phe⁷ and Met-enkephalin-Arg⁶-Gly⁷-Leu⁸, all of which are derived from proenkephalin A. The third group comprises α- and β-neoendorphin and dynorphin. These peptides have Leu-enkephalin structure at the N-terminus and are derived from the common precursor, proenkephalin B (or prodynorphin).
