1）アラキドン酸代謝物による内皮障害

Endothelial cell in a microcirculation from a viewpoint of arachidonic acid cascade.

　プロスタグランディン（PG）は1935年に発見されて以来膨大な研究が蓄積され，primary PGに続いてトロンボキサン（TX），PGI₂，ロイコトリエン（LT）などが発見され，アラキドン酸カスケードの全容が解明されてきた。血小板活性化因子（platelet-activating factor：PAF）は1972年にBenveniste^1）によりその存在が証明され，Hanahan^2）により構造決定されて以来，好中球活性化作用，平滑筋収縮作用や内皮透過性亢進などさまざまな生理活性を有する炎症性メディエーターとして認識されるに至っている。従来，PAFはアラキドン酸カスケードとは無関係な物質とされてきたが，その前駆物質がアラキドン酸と同一物質であることが解明され，広い意味でPAFはアラキドン酸関連物質であることが判明した。さらに近年，PAFとアラキドン酸代謝産物との間に複雑な相互作用が存在することが指摘されている。

　まず種々の刺激によりホスホリパーゼ（PL）A₂，Cの活性が上昇すると，おのおのホスファチジルコリンからリゾホスファチジルコリンとアラキドン酸が，またホスホイノソチドからジアシルグリセロールとイノシトールリン酸とが産生される。このジアシルグリセロールにジアシルグリセロール・リパーゼが作用してモノアシルグリセロールとアラキドン酸が産生される。このアラキドン酸にシクロオキシゲナーゼが作用するとPGG₂，PGH₂となりこれにイソメラーゼ，リダクターゼやPGI₂シンセターゼが作用することによりPGs（PGI₂，PGE₂など）が，またTXシンセターゼが作用するとTXA₂が産生される。一方，アラキドン酸に5-リポキシゲナーゼが作用すると5-HPETE（hydroperoxyeicosatetraenoicacid）が産生され，これにLTA₄シンセターゼが作用するとLTA4が産生され，これがLTB₄，LTC₄，LTD₄などに代謝されていく（図1）。

I believe that the investigation of microcirculation should be conducted with regard to the interrelationship of four elements, namely, blood flow, vessel wall (endothelial cells, pericytes etc.), cellular components in the vessels (platelets, leukocytes, erythrocytes etc.) and extravascular tissues (mast cells etc.). The recent biochemical, molecular and biological advances enabled us to explain their interaction by mediating many kinds of factors. If there is any damage to the vessels, the arachidonic acid cascade will be activated followed by the production of prostaglandins. It is very important for us to take into account the arachidonic acid metabolism in endothelial cells and the cell to cell interaction in a microcirculation.
