　虚血性脳浮腫に対するアラキドン酸の関与を検討するために，ラットの中大脳動脈閉塞により局所脳虚血モデルにおいて，アラキドン酸のcyclo-oxygenase阻害剤のインドメタシンとleukotrien産生および遊離抑制作用並びにsuperoxide anion産生阻害作用をもつ塩酸アゼラスチンとがcyclo-oxygenase系代謝物であるPGE₂，TxB₂，6—keto-PGF₁α，とlipoxygenase系代謝物であるLTC₄，および脳水分含量にどのように影響を与えるかについて検討した。インドメタシン前処置によりPGE₂，TxB₂，6—keto-PGF₁αの増加は抑制されたが，脳水分含量の増加は抑制されなかった。塩酸アゼラスチン前処置によりLTC₄の増加は抑制され，閉塞6時間後により脳水分含量の増加も有意に抑制された。以上の結果から，局所脳虚血における虚血性脳浮腫の発生にはcyclo-oxygenase系よりもlipoxygenase系代謝がより関与している可能性が示唆された。

The products resulting from arachidonic acid metabolism of the both cyclo-oxygenase and lipo-xygenase pathways possess strong physiological activities, such as vasoconstriction and the enhance-ment of vascular permiability. Therefore, it is likely that these metabolites are involved in cerebral cir-culatory disturbance and the formation of brain edema in cerebral ischemia. It is reported that intracerebral injection of leukotrien B₄, C₄, and E₄ increased blood-brain barrier permeability. Thus, it is suggested that leukotrienes may induce vaso-genic cerebral edema.
