Bone and Cartilage Destruction in Rheamatoid Arthritis. Genetic factors of rheumatoid arthritis.

　関節リウマチ（RA）などの免疫が関与する疾患は，多くは遺伝因子，環境因子など複数の要因が働いて発症する。以前より，主要組織適合遺伝子複合体をはじめ幾つかの疾患感受性遺伝子が報告されていたが，最近になりゲノムワイドの関連解析（genome-wide association study：GWAS）を用いてRAの疾患感受性遺伝子の解析が進められている。ゲノム情報は疾患の成立以前から存在しており，疾患（＝結果）に対する明確な因果関係を持つことから，ゲノム情報を用いた病態の理解と創薬への応用に期待が持たれている。

　Rheumatoid arthritis（RA）is one of common autoimmune diseases. As with other complex disorders, recent genome-wide association studies（GWASs）have greatly contributed to our understanding of RA etiology. In this review, the genetic configuration of RA as revealed through GWASs is described. In addition, I discuss the pathologic mechanisms of RA as suggested by the findings of genetic and functional studies of individual RA-associated genes, and the potential use of genetic information for novel drug discovery.
