白血病幹細胞研究の展開

Perspective on leukemia stem cell research

急性骨髄性白血病幹細胞が同定されたことによって，白血病造血にも幹細胞を頂点とするヒエラルキーモデルが適応可能と考えられるようになった．白血病幹細胞は主に細胞周期の静止期にあるため通常化学療法に抵抗性で，微小残存病変や再発の原因となっていると考えられる．多くの研究者が白血病幹細胞を特異的に死滅させる治療法の開発に努めているが，幹細胞アッセイ系としての異種移植システムは必ずしも十分な効率を達成していない．白血病幹細胞研究の根幹となる免疫不全マウスの改良にも大きな期待が寄せられる．

The identification and characterization of leukemia stem cells might lead to development of more effective treatment strategies especially for acute myelogenous leukemias.　To achieve this goal, it will be necessary to overcome technical issues related to xenotransplant mouse models by improving their tolerance for human cells as well as by providing a more appropriate microenvironment for leukemia stem cells.　It is also important that the heterogeneity arising from clonal evolution or epigenetic differences in leukemia stem cells should be taken into account.
