ポリグルタミン病における転写障害

Aberrant transcriptional regulation in polyglutamine diseases

　現在までに9つの神経変性疾患の病因遺伝子において，ポリグルタミン鎖をコードするCAGリピートの伸長が発見され，伸長ポリグルタミン鎖を含む変異蛋白自体が神経細胞変性を引き起こすものと考えられている（ポリグルタミン病）。またその病態機序において，変異蛋白の核移行が重要である可能性が指摘されている。筆者らは伸長ポリグルタミン鎖が転写因子CREBに依存した遺伝子発現を阻害することを明らかにし，ポリグルタミン病の病態機序として「伸長ポリグルタミン鎖→凝集体形成→アポトーシス」という従来の考え方から「伸長ポリグルタミン鎖の核移行→特定の遺伝子群の転写障害→可逆性の核の機能障害」という考え方へのパラダイムシフトの必要性を提唱した。本稿では，ポリグルタミン病において近年，注目されている転写調節障害について概説したい。

At least nine neurodegenerative diseases are known to be caused by expanded CAG repeats encoding polyglutamine (polyQ) stretches. Although cytotoxicities of expanded polyQ stretches have been suggested, the molecular mechanisms of neurodegeneration remain unclear. We demonstrated that the nuclear translocation of mutant proteins containing expanded polyQ stretches is a prerequisite for the expression of their cytotoxicity.
