Pathology of polyglutamine disease

　ポリグルタミン病では神経細胞における核内封入体形成が，ほぼ共通の病理所見として認識されている。ヒト脳における封入体の超微形態像は一様ではなく，核の正常構造であるnuclear bodyとの相互作用も含め，種々の分子の多様な関与が示されてきている。他方，封入体形成とともに原因蛋白質の核内びまん性蓄積も，ポリグルタミン病の病態として重要であると認識された。これらを指標として提唱された各疾患の新たな病変分布は，細胞脱落に基づく従来の変性部位をはるかに越える領域に，神経細胞の機能異常が潜在している可能性を示唆している。

It is now widely accepted that the formation of neuronal intranuclear inclusions (NIIs) is a characteristic feature of the CAG-repeat diseases including Huntington's disease and dentatorubral-pallidoluysian atrophy (DRPLA). NIIs are eosinophilic round structures and vary in size up to about 4 μm. They are not always a single structure. In the case of neurons containing two NIIs per nucleus, the inclusions frequently appeared as pairs or doublets. NIIs are heterogeneous in composition, and contain a mixture of granular and filamentous structures.
