Major parkinsonian motor symptoms, i. e., bradykinesia, muscular rigidity, and resting tremor, are due to abnormalities in the motor circuit of the basal ganglia caused by loss of nigrostriatal dopamine neurons. Bradykinesia has been assumed to result from increased activity of output nuclei of the basal ganglia, comprised of the internal segment of the globus pallidus (GPi) and the pars reticulata of the substantia nigra (SNr) , leading to excessive inhibition of the thalamocortical projections.
