Possible participation of nitric oxide in ischemic cerebral injury

　1．NO合成系

　マウス大脳皮質ホモジェネートにNOを加えると可溶性guanilate cyclaseを活性化することは古くから知られており，ラット脳可溶性分画には，非常に低分子でかつヘモグロビンで抑制されるguanilatecyclase活性効果を示す物質の存在することが指摘されていた。L-arginine（D-arginineからは産生されない）を脳シナプトゾーム分画に加えるとcitrullineとNOが生成し，可溶性guanilate cyclaseが活性化されることが示された^30）。この現象は，ヘモグロビンおよびNOS阻害剤（後述）で抑制され，脳におけるNOSの存在を示した。一方，Garthwaiteらのグループも，1988年neonatal cerebeller cell cultureからNOと考えられる血管拡張因子の生成を認めている^14）。そして，1990年Radomskiらが，脳抽出物中のL-arginineからNOが生成されることを発表した^67）。NOは，血管系への作用のみならず，中枢，末梢神経系のトランスミッターとして注目されている。

　The present study aimed at examining whether ischemic brain damage might develop mediated by the postischemic overproduction of nitric oxide (NO) in the brain.

　The first study on the effects of LNA (Nω-nitro-L-arginine) on brain water content subjected to occlusion of middle cerebral artery in rats (MCAo) elucidated that repeated intraperitoneal injection of 0.01 to 1mg/kg LNA found to be effective for its prevention.
