We performed immunohistochemical studies on pleomorphic adenoma of lacrimal gland in a 22-year-old male. We examined the binding sites of 7 antibodies and 8 lectins. The cell type of pleomor-phic adenoma was divided into 3 groups : epithelial cell lining on the luminal surface of the duct, spin-dle-shaped cells of mesenchymal origin, and trans-formed cells of both types. Anti-EMA (epithelial membrane antigen) antibody and SBA (glycinemax) were specifically bound to the epithelial cells. The mesenchymal component was labelled with anti-GFAP (glial fibrillary acidic protein) anti-body and GS-2 (griffonia simplicifolia 2). The transformed cells showed cross reactivity for anti-keratin and antivimentin antibodies and for GS-1 and PNA (arachis hypogaea). The findings suggest the usefulness of lectin cytochemistry to distinguish cell types in pleomorphic adenoma. It is presumed that GS-2 recognizes the tumor-associated antigen.
