スギ花粉症に対するトラニラスト点眼液の眼誘発反応抑制効果

Suppression of ocular reactions in cedar pollinosis by tranilast ophthalmic solution

　スギ花粉症患者12例に抗原による眼誘発試験を行い，0.5％トラニラスト（N−5′）点眼液の涙液中ヒスタミン遊離抑制効果を検討した右眼にN−5′点眼液，左眼にplacebo点眼液を点眼し，10分後にスギ抗原液を点眼して，アレルギー反応を誘発した。N−5′投与眼の誘発5分および10分後の涙液中ヒスタミン量は，対照眼に比べ有意なヒスタミン遊離抑制効果がみられた。N—5′点眼液のヒスタミン遊離抑制率は，誘発5分後50％，誘発10分後63％であった。0.5％N−5′点眼液はスギ花粉症に対する有効な薬剤であると考えられた。

We evaluated the value of topical tranilast solu-tion as provocative test for Japanese cedar pol-linosis in 12 affected patients. Tranilast was in-stilled in the right eye and the left was instilled with placebo solution. Cedar antigen was instilled in both eyes 10 minutes later to induce allergic reac-tions. Histamine level in the tear fluid was signifi-cantly lower in the right than the left eyes at 5 minutes (p＜0.001) and 10 minutes (p＜0.02) after instillation. The rate of histamine release inhibition in the tranilast-treated eyes was 50% at 5 minutes and 63% at 10 minutes after provocation. The findings suggest the possible clinical usefulness of tranilast eyedrops in the prevention and treatment of Japanese cedar pollinosis.
