老年痴呆の遺伝・染色体研究

Genetics and chromosome aberrations of senile dementia

I．はじめに

　老年期にみられる精神疾患のなかでも老年痴呆は，その病因がなお不明であり，また根本的な治療方法が未だ確立されておらず老年痴呆者に対する社会的対策をも含めて大きな社会的関心のひとつになっている。わが国における最近の調査^18）では65歳以上の老人では20人にひとり，80歳以上では4人にひとりに痴呆性疾患が認められている。今年の全人口に占める65歳以上の老人の割合は10.0％の大台にのると思われるが，もしこのままの速度で人口の高齢化が進めば痴呆老人に対する社会的負担は計り知れないものになるであろう。老年期の痴呆性疾患，なかでもアルツハイマー型痴呆（AD）の病因の早急な究明が強く求められている現況といえる。本稿ではADの遺伝学的研究について述べる。ADは遺伝学的には若年発症型と晩発型では遺伝要因あるいは臨床症状で相違がみられ，またDNAの損傷，早期老化，あるいは神経細胞数の減少などとの関連についてのいくつかの知見によってもADの病因の究明になんらかの糸口が見出されることが期待される。以下，はじめにADに関する細胞遺伝学的研究を含めた遺伝学的研究について述べる。次に、加齢とDNAの修復との関連について簡単な展望を行ない，今後の研究方向について触れたい。

Abstract

Senile dementia is now a major public health problem. It affects about one in twenty of those over 65 years of age and one in four of those aged 80 or more. If present trends of change in the age-structure of the population continue, then the burden of caring for the demented elderly could limit our capacity to provide satisfactory standards of health care for every one. In this article, some aspects of senile dementia are con-sidered from a genetic perspective.
