中枢神経組織に対するカイニン酸の毒作用について

A survey concerning the effects of kainic acid on the central nervous system

　カイニン酸（kainic acid，KA）は1953年にわが国の村上信三，竹本常松^32）らにより海人草（digena simplex）より単離抽出され，その後合成された物質である。2-カルボキシ-3-カルボキシメチル-4-イソプロピルピロリジン（図1参照）という化学構造式を有し，分子量531であり，以前わが国では回虫の駆虫薬としてよく使用されていた。この物質はその化学構造が興奮性神経伝達物質であるグルタミン酸（Glu）とよく類似しておりGluのagonistと考えられている。しかし，KAがGluと同様に作用しGluレセプターに親和性を有するかどうかについては，なお問題が残っている。KAがとくに哺乳動物中枢神経系に毒性作用を有するという報告は1974年にOlneyら^27）がKAの末梢投与により，ラット視床下部弓状核を観察したのが最初である。その後，1977年頃から神経生物学領域で実験的にKA投与を行ない，中枢神経系のいろいろの領域を破壊し，形態学的^{10,18,20,21）}，電気生理学的^15,31），神経化学的^10,12），行動科学的^6,16）に研究が行なわれている。現在までにKA投与により研究されている神経領域は，視床下部弓状核を中心とする視床下部領域に始まり^19,27），網膜^3,28），線条体^5,22），海馬^{20,21,23,24,26）}，小脳^11），嗅脳^25）などである。

　Recently kainic acid (KA) has been used as a tool for scrutinizing the neuron types in the central nervous system since it has been generally accepted to act as neurotoxic agent on the neurons with glutamate receptor. In this present article we mainly described the observation of two subjects revealed in our laboratory; the one was the influence of KA (i.p. injected) on the arcuate nucleus of the rat, the other was the effect of small doses of KA into lateral ventricle upon the hippocampal formation of the rat.
