神経栄養因子に関する一連のデータは，主として発達期の齧歯類から得られている。とくに，ノックアウトマウスやトランスジェニックマウスを利用して多くの知見が報告されている。これらと似た表現型を示すヒトの遺伝性疾患では，TrkA遺伝子異常を持つ先天性無汗無痛症とRet遺伝子異常によるHirschsprung病が上げられる。しかし，いろいろの制約があることから，ヒトのとくに成熟期・老齢期の神経栄養因子に関する系統的な研究は非常に乏しい。ヒトの病変局所における神経栄養因子の動態は臨床応用の上からもきわめて重要である。ここでは，われわれが検討してきた筋萎縮性側索硬化症とニューロパチーを中心に，病変組織における発現変化について述べる。

The distribution and expression of neurotrophic factors and their receptors have been extensively studied, especially at the embryonal stage in rodents. However, knowledge is lacking as to the expression in the adult human nervous system in terms of both physiological and pathological states. Here, the expression of GDNF and its receptor (RET and GDNFR-α) was studied in the spinal cord and muscle of patients with amyotrophic lateral sclerosis (ALS) and in the nerves of patients with peripheral neuropathies.
