抗接着因子による免疫性神経疾患の治療

Treatment of neuroimmunological diseases with antibodies against adhesion molecules

　接着因子は自己免疫疾患における免疫応答において重要な働きをしている。リンパ球のhoming，抗原提示，costimulatory factorとしての役割などの他，移植免疫，腫瘍免疫などに深く関与している。免疫性神経疾患である多発性硬化症（MS）とその動物モデルである実験的自己免疫性脳脊髄炎（EAE）につき，接着因子の病態への関与と抗接着因子抗体による治療の試みにつき概説した。接着因子のなかでもICAM-1/LFA-1 pathway，VLA-4/VCAM-1 pathway，B7/CD28 pathwayなどが発症機序に関わる重要な役割を果たしており，これらの接着因子に対する抗体を用いた検討でEAEにおいて適切な投与時期，接着因子とそのリガンドの両者に対する抗体を用いることなどによってより強い抑制効果が得られている。接着分子に対する抗体はMSなどの免疫性神経疾患の治療への応用に期待が持てる。

The role of adhesion molecules in autoimmune diseases especially in multiple sclerosis (MS) and its model, experimental autoimmune encephalomyelitis (EAE) were reviewed. Adhesion molecules play important roles in many facets of immunological responses. In active MS lesions, expressions of LFA-1/ICAM-1, VLA-4/ VCAM-1 and B7-1, B7-2/CD28 were upregulated via the action of inflammatory cytokines. Soluble forms of adhesion molecules may serve as adequate indices of the disease activity.
