代表的な神経栄養因子であるニューロトロフィンはニューロンの分化誘導,生存維持といった発生過程における作用に加えて,神経伝達の調節やそれに続く活動依存性のシナプス可塑性に関与していることが明らかになってきた。本稿ではニューロトロフィン,とくにBDNFの神経伝達に対する短期,中期,長期作用とその分子基盤について概説する。ニューロトロフィンはすべてのタイムウインドウにおいて,前シナプス部および後シナプス部の分子に作用して,興奮性神経伝達を誘起または増強する。ニューロトロフィンと神経伝達物質は協調しながらフィードフォワードな機構により神経伝達の強化,固定に関与していると考えられる。
Several lines of evidences have shown the new roles of neurotrophins which involved in neural transmission and succeeding activity-dependent plasticity of synapses in addition to their “classical” neurotrophic action in the regulation of the differentiation and survival of neurons. Here we discuss : 1) acute, 2) intermediate and 3) chronic effects of neurotrophins on neural transmission and their molecular backgrounds. Neurotrophins induces and enhances excitatory neurotransmission by modulating pre-and post-synaptic mechanisms in an acute and a chronic phase.
