The Histological Typing of Tumours of the Central Nervous System (WHO, 1993) is based on histopathological findings as well as biological assessment by immunohistochemistry of tumor. The further informations regarding the molecular mechanisms of glial differentiation will provide more simple and reasonable classification for gliomas. Recent studies revealed that the p53 function was also modulated by ATM, MDM2 and pl9ARF. Moreover p73 and p51 were found to supplement the p53 function. The alteration of these p53 related molecules might be implicated in the tumorigenesis of the gliomas without p53 mutation.The loci of PTEN and DMBT1 genes were recently identified in the common deletion area on chromosome 10 of gliomas. The alteration of PTEN seems to occur in the late phase, whereas the alteration of DMBT1 does in the early phase of the tumorigenesis of gliomas. Certain viruses includingSV40 have been identified in some gliomas by means of PCR, but the role of them in the tumorigenesis still remains unclear. In the near future the genotypic characteristics of each tumor would well represent the biological features and predict the clinical outcome of the patient harboring glioma.
