挿入突然変異によって早期老化症状を呈するKlothoマウスを樹立した。多彩な老化症状は単一遺伝子の欠損に起因しており，原因遺伝子はN端にシグナル配列，C端に膜貫通ドメイン構造をもつ1014アミノ酸からなる新規の1型膜蛋白質をコードしていた。その発現は腎臓で高く，弱い発現が中枢神経系で観察された。RT-PCRによっても，骨，皮膚，胃，肺などでは発現は認められなかった。ヒト相同遺伝子は1型膜蛋白質と共に分泌型蛋白質をコードしていた。Klothoマウスは加齢に伴って発症する多くの疾患の成り立ちを解析するための重要なモデルマウスとなると期待される。同定された原因遺伝子の機能についてはレセプター，リガンドなど，幾つかの可能性が考えられるが今後の課題である。

We recently developed a novel unique transgenic mouse with multiple age-related disorders caused by an insertion mutation. The mice homozygous for the transgene show various phenotypes resembling those found in patients with premature aging syndromes; short life span, arteriosclerosis, osteoporosis, impaired oocyte maturation and spermatogenesis, soft tissue calcification, atrophy of thymus, pulmonary emphysema, degeneration of Purkinje cells, and skin atrophy. However, the mice did not show any abnormality in development. We named this mutant klotho for one of the Fates, the Greek goddess who spins the thread of life.
