48歳女性の右眼，および44歳男性の左眼に認められた2例の角膜真菌症に対して新しく開発された抗真菌剤であるketoconazoleの内服療法を試みた。第1例では1日300mgから100mgを内服し，治療57日間，総量13,000mgで眼前手動弁であった視力が矯正0.6に回復し，潰瘍も消失，淡い混濁を残して治癒した。第2例でも1日300mgから100mgを内服し，治療20日間，総臆4,700mgで，矯正0.6であった視力が矯正1.2まで改善し，極く淡い混濁を残して治癒した。両者とも薬剤の重篤な副作用は認められなかった。なお第2例より分離されたFusarium solaniに対するketoconazoleのMICを測定した結果250μg／mlであった。また同菌をウサギ角膜に接種し治療群と対照群に分け，1日1回100mgの内服を治療群に行った結果，治療群は対照群に比較して炎症の程度がやや低くなる傾向のあることがわかった。
Two patients with fungal keratitis were treated with peroral Ketoconazole, a new antifungal agent. In both cases, epithelialization and regression of the corneal ulcer started 4 or 5 days after initiation of treatment at the daily dosis of 300 mg. Clinical signs of corneal infection disappeared completely after 3 and 8 weeks respectively. Visual acuities showed marked improvement after treatment. No signs of toxicity or adverse reaction were observed in both cases after administration totalling 4,700 and 13,000 mg respectively.
Fusarium solani was isolated from one of the cases.
