DISC電気泳動法による髄液蛋白分画—正常髄液蛋白分画と分割採取髄液蛋白分画

DISC-ELECTROPHORESIS IN ACRYLAMIDE GEL OF CEREBROSPINAL FLUID PROTEINS ANALYSIS OF NORMAL CSF PROTEINS AND CSF PROTEINS OBTAINED FOR PNEUMOENCEPHALOGRAPHY

緒言

　1959年Raymondら^1）およびOrnsteinら^2）によつて開発されたポリアクリルアミド・ゲルDISC電気泳動法は従来の髄液蛋白電気泳動法に目ざましい進展をもたらした。それは優れた分離能力を有し，低蛋白濃度のものでも微量で，しかも濃縮することなしに泳動分析できる特徴をもつている故である。1939年Tiseliusによる電気泳動法の考案以来濾紙，寒天，殿粉ゲル，セルロース・アセテート膜等による支持体の研究技法が導入されたとはいえ，髄液の濃縮操作とそれに伴う必要量の髄液の採取はまぬがれないのである。しかしDISC泳動法による髄液蛋白の報告は少なく，したがつて不明の点も少なくない。未同定の分画，γ-グロブリンの高分子量分画およびデンシトメトリーについてもなお若干の問題を残している感がある。

　著者らは昭和41年3月から髄液のDISC泳動を行なつているが，今回は以上の点にも考慮を払いながら正常髄液蛋自分画について検討し，併せて分割採取髄液蛋白分画についても報告する。

1. Analysis of 47 normal cerebrospinal fluidproteins was carried out by DISC-electrophoresis.The mean value of relative concentration of eachprotein fraction in normal cerebrospinal fluid wasas follow: Pr: 7. 9%, Alb: 53. 3%, a_L.: 3.4%, a':5.5%, Tr:9.4%, β': 4.2%, γ': 13.6%, Sa₂: 2. 8%.

2. The correlation of the electrophoretic fractionsbetween cellulose acetate-electrophoresis and DISCelectrophoresis was proned and compared withnormal serum fractions by DISC-electrophoresis.

3. In the pherograms obtained at the time ofspinal tap for pneumoencephalograpy of 27 psy-chiatric or neurologic disorders, Pr increased gra-dually, Alb decreased gradually, a_L, showed lowvalue, a' increased and ,β' (τ fraction) increasedgradually, from the initial portion to the final.The increased concentration of β-globulin by cel-lulose acetate or paper electrophoresis was due to the increased concentration of τ-fraction (β') than transferrin.

4. Abnormal cases of prealbumin were shown and origin of the proteins in cerebrospinal fluid was shortly discussed.
