脳微量摘出試料のマイクロ電気泳動法によるLDHアイソザイム分析—その部位別分布と年齢変化

THE RESOLUTION OF LACTATE DEHYDROGENESE ISOENZYMES IN SMALL BRAIN SAMPLE BY MICRO DISC ELECTROPHORESIS

　分化した脳の機能を，物質レベルで捉えるためには，部位別に微量の試料をとり出し，その生物学的活性を生化学的に検討することが要求されている。近年この方面の関心は，にわかに高まつてきているが，ここで使用したポリアクリルアミドゲルを支持体とする電気泳動法についても，ミクロ化され，脳蛋白・核酸あるいは酵素などの微量分析に用いられている。著者らも，ミクロ電気泳動による乳酸脱水素酵素（LDH）アイソザイムの分析を，細胞レベルで実施することに成功した^9）。

　ところで酵素アイソザイムについては，高分子物質の生化学的活性様態を研究するに役立つているものの，そのような同一酵素の，基質に対する多様性の生物学的意義については，必ずしも十分な解明がなされているとはいえない。近年，脳中モノアミン酸化酵素の多様性が論じられているが，化学伝達機構にも関係するとされる，これら活性モノアミン分解酵素の質的多様性についての検討がなされることは，臨床精神医学からみても，興味がもたれるところである^16,17）。

The resolution of lactate dehydrogenase iso-enzymes in small amount of tissue samples obtained from various regions of rat brain was carried out by micro disc electrophoresis and ontogenesis was studied.

1. Differences of isoenzyme patterns among small regions adjacent to each other are not so evident and those among upper brain, middle brain and lower brain are recognized.

2. From the developmental point of view, the differentiation of the isozyme patterns at birth timeis more progressed in lower brain than in upper brain. Also the period to attain maturated patterns is shorter in lower brain than in upper brain.

3. Some finding suggesting one of the aging effects to the LDH isoenzymes is revealed in aged rats.
