はじめに
情動行動,記憶および意識(睡眠)の生物学的基盤の解明は,精神と身体との接点の問題であり,精神医学領域の基本的な問題である。従来,情動行動と生体アミンとの問に重要な関連のあることは示唆されてきた5)。しかし,脳は構造的にも機械的にもheterogenousなものであるため,情動行動と生体アミンとの関連について検討してゆくには,方法論的に,脳の構造および生理的機能を基盤にしつつ,微量レベルでの生化学的検討を加えなければならないと思われる。このような考えにもとづき,われわれは,Lowryの定量的組織化学法10)11)を用いて,視床下部,海馬などにおけるMonoamine Oxi—dase (MAO)の微量測定法を検討した。これらの部位の構造別レベルにおけるMAO活性,topographical di—stributionは,未だ報告されていない。ここでは,特に,定量的組織化学法によるMAO活性の測定法について検討した結果を報告し,測定例を示したい。
The purpose of our studies is to investigate the relationships between emotional behaviors and bi-ogenic amines. In this paper, procedures for the quantitative microdetermination of monoamine ox-idase in the central nervous tissues have been described.
1) Frozen-dried sections were prepared from the adult rat brains according to the Lowry's method. The tissue samples of 1-3 tig dry weight were dissected under the stereomicroscope.
2) Buffer substrate solution containg 14C-tyramine was added to the sample in a pointed microtube, mixed without warming and incubated at 38°C for 30 min. The isotopic reaction products were ex-tracted into ethyl acetate and their radioactivity was determined in the liquid scintillation counter.
3) The reliabilities of this enzyme assay system were studied.
4) Examples of the estimation of MAO activities were demonstrated in various cellular populations of hypothalamus and in each laminar structures of hippocampus.
