マウスの胃癌

GASTRIC CANCER IN MOUSE

Ⅰ．まえがき

　マウスあるいはラットの胃癌を生成しようとする試みは，過去20年以上に亘って多くの癌研究者，ことにアメリカNIHのStewartらを中心とする学派¹⁵⁾によって大きな努力がなされた．ところが，周知のように，マウスやラットの胃は堤状の隆起limiting ridgeによって判然と前後2室に分かれている．食道に直接つながる部分は前胃とよばれ，食物の貯ぞうの役をし，胃全体の2/3を占め，扁平上皮組織でうらうちされている．これに対し，のこりの部分は後胃とよばれ，これは人の胃に相当する部分であり，胃腺や幽門腺をそなえていて，ペプシンや塩酸を分泌して食物の消化にあたり，幽門部を経て，十二指腸に連なっている．

　マウスやラットを用いての実験で，胃の病変を取扱う場合には，その病変の発生母地が前胃であるか，あるいはまた後胃に属するものであるかを充分見究めてからかからねばならない．それは，前述のように前後胃の成立ちが組織学的にそれぞれ異なるからである．すなわち，前胃に発生したものは，扁平上皮癌が主であって，これらは皮膚，口唇あるいは食道にしばしばみられる癌と同様であるからである．一方腺胃に生ずる腫瘍は，腺癌が主であるが，時には，肉腫が含まれることがある．

　5mg of 4 NQO (4-Nitroquinoline 1-oxide) or 4 HAQO (4-Hydroxyaminoquinoline 1-oxide) dissolved in alcohol was given into the stomach of mouse by tube feeding. 5mg of drug was divided into ten doses and each dose was given once every week. Squamous epithelial cancer was originated at forestomach and adenocarcinoma was arised at glandular stomach when 4 NQO was administered. Adenocarcinoma was induced at glandular stomach but no pathological change was found at fore stomach in case of 4 HAQO. 4 HAQO has the tendency to attack glandualr stomach selectively. 4 HAQO is obtained by the reduction of 4 NQO. It is seemed as a proximate carcinogenic chemical to 4 NQO. 4 HAQO has no effect to the fore stomach lined with squamous eqithelium but attacks the glandular stomach covered by gastric glands. This drug and the origination of cancer will contribute to the advance of experimental cancer research.
