We investigated the efficacy of epinastine hydrochloride ophthalmic solution, an anti-allergic drug, on ocular symptoms in conjunctivitis induced by a combination of prostaglandin D2(PGD2)and histamine in guinea pigs. The topical administration of PGD2 and histamine produced a significant increase in itch-scratch response and bulbar conjunctival hyperemia compared with that of the administration of PGD2 or histamine alone. Epinastine hydrochloride ophthalmic solution significantly inhibited the itch-scratch response and the hyperemia induced by combination of PGD2 and histamine. On the other hand, olopatadine hydrochloride ophthalmic solution significantly inhibited only the hyperemia induced by combination of PGD2 and histamine, but did not affect the itch-scratch response. These results suggest that epinastine hydrochloride ophthalmic solution may be useful in the treatment of conjunctivitis induced by a combination of histamine and PGD2.
