With a few exceptions in monogenic autoimmune diseases, most of the autoimmune diseases such as rheumatoid arthritis are multifactorial, where both genetic and environmental factors are involved in the pathogenesis. Recent genome-wide association studies (GWASs) for autoimmune diseases have revealed that multiple common variants with moderate risk are associated with disease susceptibility. Some of these genetic variants are shared by different diseases, while other genes contribute to a specific disease, suggesting a combination of genetic factors may determine an individual's susceptibility to a disease. In addition, advances in whole genome sequencing technology are opening up the era of rare variants that may substantially influence the susceptibility to disease in individuals.
