Genome biomarkers gave acceptable explanation about individual profile of efficacy and toxicity with anti-cancer therapy. One is germ-line polymorphism mainly as predictive marker for toxicity and another is somatic mutation as predictive marker for efficacy. Developing molecular targeted agents, only somatic mutations tend to be emphasized. However, germ-line polymorphisms should be considered in the future.
