 The effects of chloramphenicol succinate applied to the round window niche were investigated by observing the various electrical phenomena in the cochlea of guinea-pigs. Using a microelectrode, the endolymphatic DC potential, cochlear microphonics and the intracellular negative potential in the organ of Corti were recorded from the basal or third turn of the cochlea.

 The marked decline of the DC potential gradient at the reticular layer based on the earlier decline of the endolymphatic potential and the subsequent decrease of the intracellular negative potential was always observed with a marked loss of the cochlear microphonics.

 The depressing effects of the drug upon the various potentials were irreversible and progressive. Among of the various electrical phenomena in the cochlea, the endolymphatic potential began to decline at first. The appearance of the drug effect was much more delayed in the third turn than in the basal turn.

 Furthermore, regarding to the origin of the deleterious effect upon the potentials, the possibility on participation of the osmotic pressure of the drug solution was discussed.
